    Scientists showed that pulmonary infection with Mycobacterium tuberculosis, but not Listeria monocytogenes, induced robust lymph node expansion of a highly activated population of pathogen-specific regulatory T (Treg) cells from the pre-existing pool of thymically derived Treg cells. [Immunity]

    Immunity to Nippostrongylus brasiliensis reinfection requires pulmonary CD4+ T-cell responses. Investigators examined whether secondary lymphoid recruited or pre-existing lung CD4+ T-cell populations coordinated this immunity. [Mucosal Immunol] Abstract

    Inflammation Promotes Airway Epithelial ATP Release via Calcium-Dependent Vesicular Pathways
    Scientists investigated links between cystic fibrosis (CF)-associated airway inflammation and airway epithelial ATP release. Primary human bronchial epithelial (HBE) cells isolated from CF lungs exhibited enhanced IL-8 secretion after 6-11 days, but not 28-35 days, in culture, relative to normal HBE cells.     Chemokine Localization in Bronchial Angiogenesis
    Scientists analyzed left lung bronchoalveolar lavage fluid and left bronchus homogenates after left pulmonary artery ligation in rats, immediately after the onset of ischemia, 6 hours and 24 hours later. Additionally, they tested the effectiveness of dexamethasone on decreasing inflammation and angiogenesis at early and late stages. [PLoS One] Full Article

    Reduced Chromosome Aberration Complexity in Normal Human Bronchial Epithelial Cells Exposed to Low-LET γ-Rays and High-LET α-Particles
    Scientists exposed normal human bronchial lung epithelial cells in vitro to 0.5 and 1 Gy low-linear energy transfer (LET) γ-rays and a low fluence of high-LET α-particles and assayed for chromosome aberrations in premature chromosome condensation spreads by 24-color multiplex-fluorescence in situ hybridization. Both simple and complex aberrations were induced in a LET and dose-dependent manner. [Int J Radiat Biol]

    In-Cell Western™ Detection of Organic Cation Transporters in Bronchial Epithelial Cell Layers Cultured at an Air-Liquid Interface on Transwell® Inserts
    Researchers designed an in-cell Western™ assay for measuring levels of the different organic cation transporters (OCT) subtypes in intact layers of the human bronchial epithelial Calu-3 cell line cultured at an air-liquid interface on Transwell® inserts. The protocol was subsequently validated by exposing cell layers to compounds known to cause variations in OCT expression. [J Pharmacol Toxicol Methods] Abstract

    Rhythmic Pressure Waves Induce Mucin5AC Expression via an EGFR-Mediated Signaling Pathway in Human Airway Epithelial Cells
    Researchers tested the effect and mechanism of rhythmic pressure waves on mucin5AC (MUC5AC) expression in cultured human bronchial epithelial cells. Compared with the relevant controls, the transcriptional level of MUC5AC and the protein expressions of MUC5AC, the phospho-epidermal growth factor receptor, phospho-extracellular signal-related kinase, and phospho-Akt were all significantly increased after mechanical stimulation.     Gene Amplification of the Histone Methyltransferase SETDB1 Contributes to Human Lung Tumorigenesis
    Researchers showed that the histone methyltransferase SETDB1 undergoes gene amplification in non-small and small lung cancer cell lines and primary tumors. The existence of additional copies of the SETDB1 gene in these transformed cells is associated with higher levels of the corresponding mRNA and protein. [Oncogene] Abstract

    Pulmonary Fibroblasts Induce Epithelial Mesenchymal Transition and Some Characteristics of Stem Cells in Non-Small Cell Lung Cancer
    Fibroblasts isolated from surgical exploration were co-cultured with human lung adenocarcinoma cell lines. Expression levels of myofibroblast markers were higher in cancer associated fibroblasts than lung normal fibroblasts within five passages in the absence of continuing interaction with carcinoma cells.     Ascorbic Acid and a Cytostatic Inhibitor of Glycolysis Synergistically Induce Apoptosis in Non-Small Cell Lung Cancer Cells
    Scientists compared the effects of ascorbic acid (AA) and combinations of AA with the glycolysis inhibitor 3-(3-pyridinyl)-1-(4-pyridinyl)-2-propen​-1-one(3-PO) on the viability of three non-small cell lung cancer (NSCLC) cell lines to the effects on an immortalized lung epithelial cell line. AA concentrations of 0.5 to 5 mM caused a complete loss of viability in all NSCLC lines compared to a <10% loss of viability in the lung epithelial cell line. [PLoS One] Full Article

    Interleukin-22 Is Frequently Expressed in Small- and Large-Cell Lung Cancer and Promotes Growth in Chemotherapy-Resistant Cancer Cells
    Investigators analyzed the cellular effects of interleukin-22 (IL-22) on lung carcinoma cell lines and the prognostic impact of IL-22 tissue expression in lung cancer patients. They found the IL-22 receptor 1 to be expressed in six of seven lung cancer cell lines. [J Thorac Oncol] Abstract
