Researchers Identify ‘Achilles Heel’ in Metabolic Pathway that Could Lead to New Lung Cancer Treatments
Researchers have found an “Achilles heel” in a metabolic pathway crucial to stopping the growth of lung cancer cells. At the heart of this pathway lies peroxisome proliferation-activated receptor gamma (PPARγ), a protein that regulates glucose and lipid metabolism in normal cells. Researchers demonstrated that by activating PPARγ with antidiabetic drugs in lung cancer cells, they could stop these tumor cells from dividing. IL-27 and Type 2 Immunity in Asthmatic Patients: Association with Severity, CXCL9, and Signal Transducer and Activator of Transcription Signaling
Investigators evaluated IL-27 expression in human asthma alone and in combination with type 2 immunity to determine the relationship to disease severity and CXCL9 expression. They also modeled these interactions in vitro in human bronchial epithelial cells. [J Allergy Clin Immunol] Full Article

D Prostanoid Receptor 2 (Chemoattractant Receptor-Homologous Molecule Expressed on TH2 Cells) Protein Expression in Asthmatic Patients and Its Effects on Bronchial Epithelial Cells
Researchers investigated the bronchial submucosal D prostanoid receptor 2 expression in asthmatic patients and healthy control subjects and explored its functional role in epithelial cells. [J Allergy Clin Immunol]

Src-Mediated EGF Receptor Activation Regulates Ozone-Induced Interleukin 8 Expression in Human Bronchial Epithelial Cells
Scientists examined the role of epidermal growth factor (EGF) receptor activation in ozone-induced expression of the chemokine interleukin 8 in human bronchial epithelial cells. [Environ Health Perspect]

High Basal Expression of Interferon-Stimulated Genes in Human Bronchial Epithelial (BEAS-2B) Cells Contributes to Influenza A Virus Resistance
Investigators compared replication kinetics of human H1N1 (A/USSR/77) influenza A viruses (IAVs) in normal primary human bronchial epithelial and two commonly used respiratory epithelial cell lines namely BEAS-2B and A549 cells. They found that IAV replication was distinctly poor in BEAS-2B cells in comparison with normal primary human bronchial epithelial, A549 and Madin-Darby canine kidney cells. [PLoS One] Full Article

Novel Peptide for Attenuation of Hyperoxia-Induced Disruption of Lung Endothelial Barrier and Pulmonary Edema via Modulating Peroxynitrite Formation
Researchers found that exposure of pulmonary artery endothelial cells to hyperoxia (95% oxygen and 5% CO2) for 48 hours resulted in disruption of monolayer barrier integrity in a manner of two phases, and apoptosis occurred in the second phase. [J Biol Chem] Abstract | Full Article

Identification of a Proximal Progenitor Population from Murine Fetal Lungs with Clonogenic and Multilineage Differentiation Potential
Using a Nkx2-1mCherry reporter mouse, researchers have identified and captured Nkx2-1-expressing lung progenitor cells from the proximal lung epithelium during fetal development. These cells formed clonal spheres in semisolid culture that could be maintained in vitro and demonstrated self-renewal and expansion capabilities over multiple passages. [Stem Cell Rep] Full Article

Specific Expression of OATPs in Primary Small Cell Lung Cancer (SCLC) Cells as Novel Biomarkers for Diagnosis and Therapy
The expression of organic anion transporting polypeptides (OATPs) was elucidated in cell lines from small cell lung cancer (SCLC) and lung carcinoids and in paraffin-embedded samples from primary and metastatic SCLCs. Scientists found a strong relationship between OATP expression and the origin of the cells, as cells from primary or metastatic SCLC and carcinoid tumors differ with respect to OATP levels. [Cancer Lett] Abstract

Waltonitone Induces Apoptosis through Mir-663-Induced Bcl-2 Downregulation in Non-Small Cell Lung Cancer
Investigators treated H460 and H3255 lung cancer cells using different concentration of waltonitone. They observed that waltonitone inhibited cell viability and induced apoptosis in a concentration dependent manner, with upregulation of caspase-3 cleavage. They also observed upregulation of miR-663, a potential tumor suppressor, after waltonitone treatment. [Tumor Biology] Abstract

Pulmonary Vascular Changes in Asthma and COPD
The involvement of residential cells such as endothelial cells, smooth muscle cells and pulmonary fibroblasts, all appear to have a crucial role in the progression of vascular inflammation and remodelling. The regulatory abnormalities, growth factors and mediators implicated in the pulmonary vascular changes of asthma and chronic obstructive pulmonary disease (COPD) subjects and potential therapeutic targets are reviewed by the authors.
