Cellenkos Receives FDA Fast Track Designation for CK0803 in Amyotrophic Lateral Sclerosis (ALS)

[Cellenkos, Inc. (PR Newswire)] Cellenkos, Inc. announced that the US FDA has granted Fast Track designation to CK0803 for the treatment of Amyotrophic Lateral Sclerosis (ALS). CK0803 is an investigational, allogeneic, cord blood-derived Treg cell therapy engineered to express high levels of CXCR3 and CD11a.

T Cell Fate Is Dictated by Different Antigen-Presenting Cells in Response to Dietary Versus Gut Epithelial Self-Antigen

[Immunity] Researchers compared ovalbumin (OVA)-specific T cell fates using mice fed OVA, or expressing secreted, cytosolic (c), or transmembrane epithelial OVA. BATF3+ antigen-presenting cells were indispensable for CD4+ T cell proliferation only in cOVA mice, yet they drove Treg cell differentiation across all epithelial OVA models.

CD137 Agonists in Mice Suppress Germinal Center Responses by Increasing the Immunosuppressive Activity of Follicular Regulatory T-Cells

[EMBO Journal] Investigators showed that agonistic anti-CD137 monoclonal antibodies target multiple CD137-expressing cell types, including activated CD8 T-cells, Tregs, follicular Tregs, and follicular dendritic cells.

CD97/ADGRE5 Attenuates the Induction of Adaptive Type 2 Immune Responses in Allergic Asthma

[Nature Communications] Scientists investigated the function of CD97/ADGRE5, expressed in mouse and human immune and lung epithelial cells, in allergic asthma. A single CD97 antibody treatment before allergen sensitization worsens allergic responses, highlighting a role for CD97 in early immune regulation.

Unconventional Role of Hydroxymethylglutaryl-CoA Synthase 1 in Driving Pathogenic T17 Cell Immunity and Autoimmune Diseases

[Science Advances] Scientists demonstrated that HMGCS1, a cholesterol biosynthesis precursor enzyme, was highly induced by inflammatory cytokines and preferentially expressed by pathogenic T helper 17 cells in vitro and in vivo.

Reduced SRSF1 Expression Correlates with LCK Exon Skipping and Impaired Treg Induction in Immune Thrombocytopenia

[Blood] Researchers showed that CD4+ naive T cells from patients with immune thrombocytopenia exhibit diminished T cell receptor signaling and impaired in vitro Treg induction relative to healthy controls.

Metabolic Switch: Lymphocytes and the Onset of T Cell-Mediated Autoimmunity

[Cellular & Molecular Immunology] The authors highlight immunometabolism as a key mechanism in autoimmunity. Immunometabolic pathways represent a new avenue for precision immunotherapy, although challenges persist in targeting cells specifically without systemic toxicity.

H3.3 Chaperone Hira Primes the Effector Program and Function of Regulatory T Cells

[Science Advances] Scientists showed that the histone variant H3.3 is enriched in tissue Tregs compared to splenic Tregs and its chaperone Hira is a critical regulator of Treg effector program.

Epigenetic Regulation of B Cell Tolerance and Dysfunction in Autoimmune Disease

[Nature Reviews Immunology] Investigators describe the molecular mechanisms by which multiple epigenetic layers, encompassing DNA methylation, histone modification, chromatin remodelling, non-coding RNA regulation and RNA modification, collectively regulate normal B cell function and, when dysregulated, contribute to autoimmune pathogenesis.

Liver Regeneration CRISPR Screen Identifies E3 Ligase NEURL1B Regulates Hepatocyte Mitosis by Destabilizing Microtubule Organizing Centers

[Gastroenterology] To understand how hepatocytes cope with mitotic challenges, the authors established a high-quality in vivo CRISPRi screen in regenerating mouse liver. By cross-referencing with gene essentiality data in cell lines, they identified genes required specifically by hepatocytes for proliferation.

CXCR4-Targeted Dual Softener for Inducing Senescence Therapy in Hepatic Fibrosis

[Proceedings of the National Academy of Sciences of the United States of America] Scientists developed an activated hepatic stellate cells (aHSCs)-targeted nanoplex that activates the stimulator of interferon genes pathway to induce senescence in aHSCs, while promoting endogenous immune surveillance-mediated clearance of aHSCs.

Hepatocellular Carcinoma Organoids Enable Drug Discovery and Combination Therapy across Heterogeneous Tumors

[Cell Reports] Researchers developed a patient-derived liver cancer organoid platform for high-throughput drug screening, identifying promising drug candidates and combination therapies across heterogeneous tumors.
