Reciprocal Interactions between the Gut Microbiome and Mammary Tissue Mast Cells Promote Metastatic Dissemination of HR+ Breast Tumors

Scientists investigated whether cellular changes occurring in normal mammary tissues, prior to tumor initiation and in response to dysbiosis, enhanced dissemination of hormone receptor (HR)+ tumors.

ABL Kinases Regulate Translation in HER2+ Cells Through Y-Box-Binding Protein 1 to Facilitate Colonization of the Brain

Mechanistically, Abelson (ABL) kinases phosphorylated the RNA-binding protein Y-box-binding protein 1 (YB-1). ABL kinase inhibition disrupted binding of YB-1 to the ERBB2 mRNA and impaired translation, leading to a profound decrease in HER2 protein levels.

A High-Throughput Drug Screen Reveals Means to Differentiate Triple-Negative Breast Cancer

Using a high-throughput imaging-based reporter drug screen with 9501 compounds, investigators identified three polo-like kinase 1 inhibitors as major inducers of estrogen receptor α protein expression and downstream activity in TNBC cells.

Human WDR5 Promotes Breast Cancer Growth and Metastasis via KMT2-Independent Translation Regulation

Scientists found that knockdown of WDR5 in breast cancer cells independently impaired their tumorigenic as well as metastatic capabilities.

The Drug Efflux Pump MDR1 Promotes Intrinsic and Acquired Resistance to PROTACs in Cancer Cells

Using proteomic analysis, the authors found intrinsic and acquired resistance mechanisms to proteolysis-targeting chimeras (PROTACs) in cancer cell lines mediated by greater abundance or production of the drug efflux pump MDR1.

Inhibitory Effect of the Novel Tyrosine Kinase Inhibitor DCC-2036 on Triple-Negative Breast Cancer Stem Cells Through AXL-KLF5 Positive Feedback Loop

Researchers found that the small-molecule tyrosine kinase inhibitor DCC-2036 suppressed TNBC stem cells by inhibiting the tyrosine kinase AXL and the transcription factor KLF5.

GASP1 Enhances Malignant Phenotypes of Breast Cancer Cells and Decreases Their Response to Paclitaxel by Forming a Vicious Cycle with IGF1/IGF1R Signaling Pathway

Scientists demonstrated that GASP1 was highly expressed in breast cancers, and patients harboring altered GASP1 showed a worse prognosis than those with wild-type GASP1.

Combining Organoid Models with Next-Generation Sequencing to Reveal Tumor Heterogeneity and Predict Therapeutic Response in Breast Cancer

Investigators found that the histological characteristics of organoids were consistent with original lesions and recapitulated their heterogenicity. In addition, the NGS results showed that PIK3CA and TP53 genes had detrimental mutations.

Senescence and Cancer — Role and Therapeutic Opportunities

Researchers discuss the major mechanisms involved in the antitumorigenic functions of senescent cells, and then consider the cell-intrinsic and cell-extrinsic factors that participate in their switch towards a tumour-promoting role.

The RING Finger Protein Family in Health and Disease

The authors summarize the potential contribution of dysregulated ring finger (RNF) proteins, except for tripartite-motif family, to the pathogenesis of some diseases, including cancer, autoimmune diseases, and neurodegenerative disorder.
