Up-Regulation of C/EBPα Inhibits Suppressive Activity of Myeloid Cells and Potentiates Antitumor Response in Mice and Cancer Patients

[Clinical Cancer Research] The authors conducted a Phase I trial in 36 patients with hepatocellular carcinoma who were given therapeutic C/EBPα saRNA (MTL-CEBPA) as either neoadjuvant or adjuvant treatment. MTL-CEBPA treatment in those patients caused a marked decrease in peripheral blood monocytic myeloid-derived suppressor cell numbers.

Hematopoietic Upstream Stimulating Factor 1 Deficiency Is Associated with Increased Atherosclerosis Susceptibility in LDL Receptor Knockout Mice

[Scientific Reports] Scientists tested whether selective hematopoietic upstream stimulatory factor 1 (USF1) deficiency could also beneficially impact the development of atherosclerosis. Absence of USF1 function in bone marrow-derived cells was associated with exacerbated blood leukocyte and peritoneal leukocyte lipid loading and an increased atherosclerosis susceptibility.

Selective Deletion of Human Leukocyte Antigens Protects Stem Cell-Derived Islets from Immune Rejection

[Cell Reports] Scientists used genetic engineering to create human PSCs in which the majority of the polymorphic human leukocyte antigens, the main drivers of allogeneic rejection, were deleted.

How the Immune System Shapes Atherosclerosis: Roles of Innate and Adaptive Immunity

[Nature Reviews Immunology] The authors review key molecular mechanisms of immune cell activation implicated in modulating atherogenesis and provide an update on the contributions of innate and adaptive immune cell subsets in atherosclerosis.

Human T Cells Engineered with a Leukemia Lipid-Specific TCR Enables Donor-Unrestricted Recognition of CD1c-Expressing Leukemia

[Nature Communications] Scientists showed that human T cells engineered to express an mLPA-specific TCR, targeted diverse CD1c-expressing leukemia blasts in vitro and significantly delayed the progression of three models of leukemia xenograft in NSG mice, an effect that was boosted by mLPA-cellular immunization.

Autophagy Modulates Endothelial Junctions to Restrain Neutrophil Diapedesis during Inflammation

[Immunity] Researchers demonstrated that endothelial cells (ECs) of venules in inflamed tissues exhibited a robust autophagic response that was aligned temporally with the peak of neutrophil trafficking and was strictly localized to EC contacts.
