HT-mediated%20extravasation%20was%20significantly%20reduced%20by%20pharmacological%20inhibition%20of%20the%205-HT2A%20receptor%20subtype%2C%20or%20with%20antagonism%20or%20deletion%20of%20TRPV4%2C%20consistent%20with%20functional%20interaction%20between%205-HT%20receptors%20and%20TRPV4.%20Inhibition%20of%20receptors%20for%20the%20neuropeptides%20substance%20P%20%28SP%29%20or%20calcitonin%20gene-related%20peptide%20%28CGRP%29%20diminished%205-HT-induced%20plasma%20extravasation.%20Supporting%20studies%20assessing%20treatment%20of%20HUVEC%20with%205-HT%2C%20CGRP%2C%20or%20SP%20was%20associated%20with%20ERK%20phosphorylation.%20Exposure%20to%20the%20TRPV4%20activator%20GSK1016790A%2C%20but%20not%205-HT%2C%20increased%20intracellular%20Ca2%2B%20in%20these%20cells.%20However%2C%205-HT%20pre-treatment%20enhanced%20GSK1016790A-mediated%20Ca2%2B%20signaling%2C%20consistent%20with%20sensitization%20of%20TRPV4.%20The%20functional%20interaction%20was%20further%20characterized%20in%20HEK293%20cells%20expressing%205-HT2A%20to%20reveal%20that%20TRPV4%20enhances%20the%20duration%20of%205-HT-evoked%20Ca2%2B%20signaling%20through%20a%20PLA2%20and%20PKC-dependent%20mechanism.%20In%20summary%2C%20this%20study%20demonstrates%20that%20TRPV4%20contributes%20to%205-HT2A-induced%20plasma%20extravasation%20in%20the%20airways%20and%20upper%20GI%20tract%2C%20with%20evidence%20supporting%20a%20mechanism%20of%20action%20involving%20SP%20and%20CGRP%20release.%22%2C%22date%22%3A%222021-04-15%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41374-021-00593-7%22%2C%22ISSN%22%3A%221530-0307%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41374-021-00593-7%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-04-15T16%3A16%3A02Z%22%7D%7D%5D%7D
