ophenotypically%20indistinguishable%20AML%20can%20be%20influenced%20by%20their%20different%20cells%20of%20origin%2C%20corresponding%20evidence%20in%20human%20disease%20is%20still%20largely%20lacking.%20By%20combining%20prospective%20disease%20modeling%20using%20highly%20purified%20human%20hematopoietic%20stem%20or%20progenitor%20cells%20with%20retrospective%20deconvolution%20study%20of%20leukemia%20stem%20cells%20%28LSCs%29%20from%20primary%20patient%20samples%2C%20we%20identified%20human%20hematopoietic%20stem%20cells%20%28HSCs%29%20and%20common%20myeloid%20progenitors%20%28CMPs%29%20as%20two%20distinctive%20origins%20of%20human%20AML%20driven%20by%20Mixed%20Lineage%20Leukemia%20%28MLL%29%20gene%20fusions%20%28MLL-AML%29.%20Despite%20LSCs%20from%20either%20MLL-rearranged%20HSCs%20or%20MLL-rearranged%20CMPs%20having%20a%20mature%20CD34%5Cu2212%5C%2Flo%5C%2FCD38%2B%20immunophenotype%20in%20both%20a%20humanized%20mouse%20model%20and%20primary%20patient%20samples%2C%20the%20resulting%20AML%20cells%20exhibited%20contrasting%20responses%20to%20chemotherapy.%20HSC-derived%20MLL-AML%20was%20highly%20resistant%20to%20chemotherapy%20and%20expressed%20elevated%20amounts%20of%20the%20multispecific%20anion%20transporter%20ABCC3.%20Inhibition%20of%20ABCC3%20by%20shRNA-mediated%20knockdown%20or%20with%20small-molecule%20inhibitor%20fidaxomicin%2C%20currently%20used%20for%20diarrhea%20associated%20with%20Clostridium%20difficile%20infection%2C%20effectively%20resensitized%20HSC-derived%20MLL-AML%20toward%20standard%20chemotherapeutic%20drugs.%20This%20study%20not%20only%20functionally%20established%20two%20distinctive%20origins%20of%20human%20LSCs%20for%20MLL-AML%20and%20their%20role%20in%20mediating%20chemoresistance%20but%20also%20identified%20a%20potential%20therapeutic%20avenue%20for%20stem%20cell%5Cu2013associated%20treatment%20resistance%20by%20repurposing%20a%20well-tolerated%20antidiarrhea%20drug%20already%20used%20in%20the%20clinic.%5CnChemoresistant%20human%20CD34%5Cu2212%5C%2Flo%5C%2FCD38%2B%20MLL%20leukemic%20stem%20cells%20originate%20from%20HSCs%20and%20can%20be%20resensitized%20by%20targeting%20ABCC3%20with%20fidaxomicin.%5CnChemoresistant%20human%20CD34%5Cu2212%5C%2Flo%5C%2FCD38%2B%20MLL%20leukemic%20stem%20cells%20originate%20from%20HSCs%20and%20can%20be%20resensitized%20by%20targeting%20ABCC3%20with%20fidaxomicin.%22%2C%22date%22%3A%222021%5C%2F02%5C%2F24%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1126%5C%2Fscitranslmed.abc4822%22%2C%22ISSN%22%3A%221946-6234%2C%201946-6242%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fstm.sciencemag.org%5C%2Fcontent%5C%2F13%5C%2F582%5C%2Feabc4822%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-02-25T17%3A14%3A04Z%22%7D%7D%5D%7D
