ndings%2C%20in%20addition%20to%20the%20adoptive%20transfer%20studies%2C%20validated%20the%20role%20of%20immunosuppressive%20CD11b%2BGr1%2B%20myeloid%20cells%20in%20promoting%20ovarian%20cancer.%20Mechanistic%20investigations%20showed%20that%20ID8%20tumor%20cells%20and%20their%20microenvironment%20produced%20both%20recruitment%20and%20regulatory%20factors%20for%20immunosuppressive%20CD11b%2BGr1%2B%20myeloid%20cells.%20CD11b%2BGr1%2B%20myeloid%20cells%20primed%20by%20ID8%20tumors%20showed%20increased%20immunosuppressive%20marker%20expression%20and%20acquired%20an%20energetic%20metabolic%20phenotype%20promoted%20mainly%20by%20increased%20oxidative%20phosphorylation%20fueled%20by%20glutamine.%20Inhibiting%20the%20glutamine%20metabolic%20pathway%20reduced%20the%20increased%20oxidative%20phosphorylation%20and%20decreased%20immunosuppressive%20markers%20expression%20and%20function.%20Dihydrolipoamide%20succinyl%20transferase%20%28DLST%29%2C%20a%20subunit%20of%20%5Cu03b1-KGDC%20in%20the%20TCA%20cycle%2C%20was%20found%20be%20the%20most%20significantly%20elevated%20gene%20in%20tumor%20primed%20myeloid%20cells.%20Inhibition%20of%20DLST%20reduced%20oxidative%20phosphorylation%2C%20immunosuppressive%20marker%20expression%2C%20and%20function%20in%20myeloid%20cells.%5CnConclusion%5CnOur%20study%20shows%20that%20the%20ovarian%20cancer%20microenvironment%20can%20regulate%20the%20metabolism%20and%20function%20of%20immunosuppressive%20CD11b%2BGr1%2B%20myeloid%20cells%20and%20modulate%20its%20immune%20microenvironment.%20Targeting%20glutamine%20metabolism%20via%20DLST%20in%20those%20immunosuppressive%20myeloid%20decreased%20their%20activity%2C%20leading%20to%20a%20reduction%20in%20the%20immunosuppressive%20tumor%20microenvironment.%20Thus%2C%20targeting%20glutamine%20metabolism%20has%20the%20potential%20to%20enhance%20the%20success%20of%20immunotherapy%20in%20ovarian%20cancer.%22%2C%22date%22%3A%22June%2016%2C%202021%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1016%5C%2Fj.molmet.2021.101272%22%2C%22ISSN%22%3A%222212-8778%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.sciencedirect.com%5C%2Fscience%5C%2Farticle%5C%2Fpii%5C%2FS2212877821001174%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-06-21T17%3A07%3A29Z%22%7D%7D%5D%7D
