Blocking Senescence and Tolerogenic Function of Dendritic Cells Induced by γδ Treg Cells Enhances Tumor-Specific Immunity for Cancer Immunotherapy

[Journal Of Immunotherapy of Cancer] Impaired tumor antigen HER2-specific recognition and immune response of senescent denderitic cells (DC) induced by γδ Treg cells were explored in vitro and in vivo in humanized mouse models. In addition, the DC-based HER2 tumor vaccine immunotherapy in breast cancer models was performed to explore the enhanced antitumor immunity via prevention of DC senescence through blockages of STAT3 and PD-L1 signaling.

Targeting Pathogenic CD8+ Tissue-Resident T Cells with Chimeric Antigen Receptor Therapy in Murine Autoimmune Cholangitis

[Nature Communications] Treatment of Il12b-/-Il2ra-/- mice mice with PD-1-targeting CAR-T cells selectively depleted liver CD8+ memory T (Trm) cells and alleviated autoimmune cholangitis. Our work highlights the pathogenic role of CD8+ Trm cells and the potential therapeutic usage of PD-1-targeting CAR-T cells.

POLQ Inhibition Attenuates the Stemness and Ferroptosis Resistance in Gastric Cancer Cells via Downregulation of Dihydroorotate Dehydrogenase

[Cell Death & Disease] The authors reported that polymerase theta (POLQ), a widely conserved DNA polymerase, positively regulated stem cell-like characteristics of gastric cancer (GC) cells. Knockdown of POLQ suppressed the stemness of GC cells in vitro and in vivo.

p53 Promotes Revival Stem Cells in the Regenerating Intestine after Severe Radiation Injury

[Nature Communications] Scientists performed single-cell RNA-seq and lineage tracing experiments and demonstrated that transient activation of p53 was required to properly reprogram damaged epithelial cells in response to severe radiation injury to promote tissue regeneration.

Activation of the YY1-UGT2B7 Axis Promotes Mammary Estrogen Homeostasis Dysregulation and Exacerbates Breast Tumor Metastasis

[Drug Metabolism And Disposition] Researchers demonstrated that up-regulation of YY1 expression was closely associated with breast cancer metastasis, and that high YY1 expression could promote the migratory invasive ability of breast cancer cells.

Conductive Hyaluronic Acid/Deep Eutectic Solvent Composite Hydrogel as a Wound Dressing for Promoting Skin Burn Healing under Electrical Stimulation

[Advanced Healthcare Materials] The authors introduced a double-network conductive eutectogel, simulating the conductivity and mechanical strength of natural human skin. When combined with exogenous electrical stimulation, the conductive eutectogel exhibited the ability to reduce inflammation, stimulate cell proliferation and migration, promote collagen deposition and angiogenesis, and facilitate skin tissue remodeling.
