0stress%20and%20serine%20protease%20activity%20in%20enterocytes%20and%20its%20impact%20on%20intestinal%20functions.the%20impact%20of%20ER%20stress%20induced%20by%20Thapsigargin%20on%20serine%20protease%20secretion%20was%20studied%20using%20either%20human%20intestinal%20cells%20lines%20or%20organoids.%20Moreover%2C%20treating%20human%20intestinal%20cells%20with%20Protease-Activated%20Receptors%20antagonists%20allows%20the%20investigation%20of%20the%20ER%20stress-resulting%20molecular%20mechanisms%20that%20induce%20proteolytic%20activity%20and%20alter%20intestinal%20epithelial%20cell%20biology.In%20our%20study%2C%20colonic%20biopsies%20from%20IBD%20patients%20exhibited%20increased%20epithelial%20trypsin-like%20activity%20associated%20with%20elevated%20ER%20stress.%20Induction%20of%20ER%20stress%20in%20human%20intestinal%20epithelial%20cells%20displayed%20enhanced%20apically%20trypsin-like%20activity.%20ER%20stress-induced%20increased%20trypsin%20activity%20destabilized%20intestinal%20barrier%20function%20by%20increasing%20permeability%20and%20by%20controlling%20inflammatory%20mediators%20such%20as%20C-X-C%20chemokine%20ligand%208%20%28CXCL8%29.%20The%20deleterious%20impact%20of%20ER%20stress-associated%20trypsin%20activity%20was%20specifically%20dependent%20on%20the%20activation%20of%20Protease-Activated%20Receptors%202%20and%204.In%20conclusion%2C%20excessive%20ER%20stress%20in%20IECs%20caused%20an%20increased%20release%20of%20trypsin%20activity%20that%2C%20in%20turn%2C%20altered%20intestinal%20barrier%20function%2C%20then%20promoting%20the%20development%20of%20inflammatory%20process.%22%2C%22date%22%3A%22February%2020%2C%202021%22%2C%22language%22%3A%22%22%2C%22DOI%22%3A%2210.1093%5C%2Fecco-jcc%5C%2Fjjab035%22%2C%22ISSN%22%3A%221873-9946%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fdoi.org%5C%2F10.1093%5C%2Fecco-jcc%5C%2Fjjab035%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-02-24T04%3A30%3A15Z%22%7D%7D%5D%7D
