ecific%20intrinsic%20tumor%20feature%20was%20broadly%20associated%20with%20sustained%20organoid%20growth.%20In%20addition%2C%20we%20performed%20phenotypic%20and%20molecular%20characterization%20of%20tumor-organoid%20pairs%20using%20immunohistochemistry%2C%20immunofluorescence%2C%20fluorescence%20in%20situ%20hybridization%2C%20and%20targeted%20sequencing.%20Morphological%20and%20immunohistochemical%20profiles%20of%20whole%20organoids%20altogether%20provided%20a%20fast%20read-out%20to%20identify%20the%20most%20promising%20ones.%20Notably%2C%20primary%20samples%20were%20associated%20with%20an%20initial%20take-rate%20of%2083%25%20%28n%3D60%5C%2F72%29%20in%20culture%2C%20with%20maintenance%20of%20cancer%20cells%20displaying%20common%20PCa%20alterations%2C%20such%20as%20PTEN%20loss%20and%20ERG%20overexpression.%20These%20cancer%20organoids%20were%20however%20progressively%20overgrown%20by%20organoids%20with%20a%20benign-like%20phenotype.%20Finally%2C%20out%20of%209%20metastasis%20samples%2C%20we%20generated%20a%20novel%20organoid%20model%20derived%20from%20a%20hormone-naive%20lung%20metastasis%2C%20which%20displays%20alterations%20in%20the%20PI3K%5C%2FAkt%20and%20Wnt%5C%2F%5Cu00df-catenin%20pathways%20and%20responds%20to%20androgen%20deprivation.%20Taken%20together%2C%20our%20comprehensive%20study%20explores%20determinants%20of%20outcome%20and%20highlights%20the%20opportunities%20and%20challenges%20associated%20with%20the%20establishment%20of%20stable%20tumor%20organoid%20lines%20derived%20from%20PCa%20patients.%22%2C%22date%22%3A%22%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%22https%3A%5C%2F%5C%2Fdoi.org%5C%2F10.1002%5C%2Fpath.5698%22%2C%22ISSN%22%3A%221096-9896%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fonlinelibrary.wiley.com%5C%2Fdoi%5C%2Fabs%5C%2F10.1002%5C%2Fpath.5698%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-05-03T21%3A27%3A15Z%22%7D%7D%5D%7D
