omain%20%28RBD%29-only%20vaccine%20fails%20to%20trigger%20long-lasting%20protective%20immunity%20if%20used%20alone%20for%20vaccination.%20To%20enhance%20antigen%20processing%20and%20cross-presentation%20in%20draining%20lymph%20nodes%20%28DLNs%29%2C%20we%20developed%20an%20interferon%20%28IFN%29-armed%20RBD%20dimerized%20by%20an%20immunoglobulin%20fragment%20%28I-R-F%29.%20I-R-F%20efficiently%20directs%20immunity%20against%20RBD%20to%20DLNs.%20A%20low%20dose%20of%20I-R-F%20induces%20not%20only%20high%20titers%20of%20long-lasting%20neutralizing%20antibodies%20%28NAbs%29%20but%20also%20more%20comprehensive%20T%20cell%20responses%20than%20RBD.%20Notably%2C%20I-R-F%20provides%20comprehensive%20protection%20in%20the%20form%20of%20a%20one-dose%20vaccine%20without%20an%20adjuvant.%20Our%20study%20shows%20that%20the%20pan-epitope%20modified%20human%20I-R-F%20%28I-P-R-F%29%20vaccine%20provides%20rapid%20and%20complete%20protection%20throughout%20the%20upper%20and%20lower%20respiratory%20tracts%20against%20a%20high-dose%20SARS-CoV-2%20challenge%20in%20rhesus%20macaques.%20Based%20on%20these%20promising%20results%2C%20we%20have%20initiated%20a%20randomized%2C%20placebo-controlled%2C%20phase%20I%5C%2FII%20trial%20of%20the%20human%20I-P-R-F%20vaccine%20%28V-01%29%20in%20180%20healthy%20adults%2C%20and%20the%20vaccine%20appears%20safe%20and%20elicits%20strong%20antiviral%20immune%20responses.%20Due%20to%20its%20potency%20and%20safety%2C%20this%20engineered%20vaccine%20may%20become%20a%20next-generation%20vaccine%20candidate%20in%20the%20global%20effort%20to%20overcome%20COVID-19.%22%2C%22date%22%3A%222021-07-15%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41422-021-00531-8%22%2C%22ISSN%22%3A%221748-7838%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41422-021-00531-8%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-07-15T18%3A10%3A53Z%22%7D%7D%5D%7D
