0place%20in%20germinal%20centers%20%28GCs%29%2C%20structures%20that%20form%20within%20secondary%20lymphoid%20organs%20in%20response%20to%20infection%20or%20immunization.%20Because%20somatic%20mutations%20accumulate%20progressively%20with%20time%2C%20the%20duration%20of%20the%20GC%20reaction%20is%20an%20important%20determinant%20of%20the%20extent%20to%20which%20antibodies%20can%20mutate%20and%20mature.%20Despite%20the%20importance%20of%20this%20parameter%2C%20our%20understanding%20of%20the%20factors%20that%20determine%20the%20duration%20of%20a%20GC%20and%20the%20timing%20of%20its%20contraction%20remains%20limited.%5CnRATIONALEIn%20addition%20to%20B%20cells%2C%20a%20minor%20but%20critical%20component%20of%20the%20GC%20reaction%20are%20T%20follicular%20helper%20%28TFH%29%20cells%20characterized%20by%20expression%20of%20chemokine%20receptor%20CXCR5%2C%20inhibitory%20receptor%20PD-1%2C%20and%20transcription%20factor%20Bcl6.%20TFH%20cells%20provide%20mitogenic%20signals%20essential%20for%20the%20proliferative%20expansion%20of%20high-affinity%20B%20cells%20and%20thus%20play%20a%20key%20role%20in%20antibody%20affinity%20maturation.%20The%20size%20of%20the%20TFH%20cell%20population%20closely%20correlates%20with%2C%20and%20likely%20determines%2C%20the%20magnitude%20of%20the%20GC%20reaction%2C%20and%20interruption%20of%20the%20signals%20provided%20by%20TFH%20cells%20to%20B%20cells%20leads%20to%20GC%20dissolution%2C%20making%20TFH%20cells%20likely%20candidates%20for%20regulators%20of%20GC%20duration.%20The%20positive%20effects%20of%20TFH%20cells%20are%20counterbalanced%20by%20a%20population%20of%20GC-resident%20T%20cells%20that%20express%20Foxp3%2C%20the%20master%20transcription%20factor%20of%20the%20regulatory%20T%20cell%20%28Treg%29%20lineage.%20The%20best%20characterized%20of%20these%20Foxp3%20populations%20are%20T%20follicular%20regulatory%20%28TFR%29%20cells%2C%20which%20arise%20through%20acquisition%20of%20a%20TFH-like%20phenotype%20by%20thymic-derived%20Tregs%20and%20control%20aspects%20of%20the%20B%20cell%20response%20ranging%20from%20B%20cell%20foreign%20and%20self-antigen%20specificity%20to%20isotype%20switching.%20Given%20the%20well-established%20role%20of%20CD4%20T%20cells%20in%20sustaining%20GCs%20and%20the%20emerging%20role%20of%20Foxp3%20in%20suppressing%20this%20reaction%2C%20we%20sought%20to%20determine%20whether%20expression%20of%20Foxp3%20by%20GC-resident%20T%20cells%20could%20also%20play%20a%20role%20in%20GC%20longevity.%5CnRESULTSImaging%20of%20the%20GC%20reactions%20in%20mice%20over%20time%20showed%20that%20the%20density%20and%20number%20of%20Foxp3%20T%20cells%20within%20these%20structures%20surged%20acutely%20in%20the%20days%20preceding%20the%20onset%20of%20GC%20contraction.%20In%20contrast%20to%20their%20early-%20or%20peak-GC%20counterparts%2C%20late-GC%20Foxp3%20T%20cells%20expressed%20high%20levels%20of%20TFH%20cell%20surface%20markers%20CXCR5%20and%20PD-1%20and%20engaged%20in%20long-lived%20dynamic%20interactions%20with%20GC%20B%20cells%20that%20resembled%20those%20observed%20for%20TFH%20cells%2C%20suggesting%20late-GC%20Foxp3%20T%20cells%20may%20differ%20in%20ontogeny%20from%20the%20canonical%20TFR%20population%20abundant%20at%20earlier%20time%20points.%20T%20cell%20receptor%20sequencing%20suggested%20that%2C%20unlike%20TFR%20cells%2C%20late-GC%20Foxp3%20T%20cells%20likely%20arise%20through%20up-regulation%20of%20Foxp3%20by%20TFH%20cells%20in%20the%20days%20immediately%20preceding%20GC%20regression.%20This%20conversion%20was%20confirmed%20by%20adoptive%20transfer%20experiments%2C%20in%20which%20transferred%20Foxp3%20na%5Cu00efve%20T%20cells%20acquired%20Foxp3%20expression%20in%20late%20but%20not%20in%20early%20GCs.%20Transcriptionally%2C%20late-GC%20Foxp3%20T%20cells%20closely%20resembled%20TFH%20cells%20while%20showing%20shifts%20in%20expression%20of%20Treg-related%20signatures%20that%20included%20loss%20of%20key%20T%20cell%20help-related%20genes.%20A%20gain-of-function%20experiment%20showed%20that%20ectopic%20Foxp3%20expression%20by%20TFH%20cells%2C%20although%20incapable%20of%20completely%20shifting%20these%20cells%20toward%20a%20TFR%20phenotype%2C%20was%20sufficient%20to%20change%20expression%20of%20Treg-related%20gene%20signatures%20and%20trigger%20GC%20contraction.%5CnCONCLUSIONOur%20data%20indicate%20that%2C%20in%20addition%20to%20canonical%20TFR%20cells%2C%20there%20exists%20a%20second%20population%20of%20Foxp3%20GC%20T%20cells%20that%20arises%20immediately%20before%20GC%20contraction%2C%20through%20the%20up-regulation%20of%20Foxp3%20and%20limited%20acquisition%20of%20Treg-like%20features%20by%20TFH%20cells.%20Functional%20experiments%20support%20a%20model%20in%20which%20the%20contraction%2C%20and%20eventual%20shutdown%2C%20of%20late-stage%20GCs%20is%20promoted%20by%20acquisition%20of%20Foxp3%20by%20this%20TFH%20cell%20population.%20These%20findings%20raise%20the%20possibility%20that%20GC%20shutdown%20is%20an%20active%20process%20rather%20than%20simply%20a%20result%20of%20the%20progressive%20consumption%20of%20antigen%20by%20GC%20B%20cells.%20Manipulating%20this%20process%20may%20provide%20an%20avenue%20toward%20extending%20GC%20lifetime%2C%20potentially%20contributing%20to%20the%20induction%20of%20highly%20mutated%20antibodies%20by%20vaccination.%20%3Cimg%20class%3D%5C%22fragment-image%5C%22%20aria-describedby%3D%5C%22F1-caption%5C%22%20src%3D%5C%22https%3A%5C%2F%5C%2Fscience.sciencemag.org%5C%2Fcontent%5C%2Fsci%5C%2F373%5C%2F6552%5C%2Feabe5146%5C%2FF1.medium.gif%5C%22%5C%2F%3E%20Download%20high-res%20image%20Open%20in%20new%20tab%20Download%20Powerpoint%20A%20surge%20in%20Foxp3%2B%20T%20cells%20precedes%20GC%20contraction.GCs%20are%20usually%20temporary%20structures%20that%20emerge%20upon%20infection%20or%20immunization%2C%20contracting%20and%20eventually%20disappearing%20upon%20cessation%20of%20antigenic%20stimulation.%20At%20their%20peak%2C%20GCs%20contain%20two%20major%20populations%20of%20T%20cells%2C%20TFH%20cells%20and%20TFR%20cells.%20Of%20these%2C%20only%20the%20latter%20population%20expresses%20the%20Treg-associated%20transcription%20factor%20Foxp3.%20We%20find%20that%20the%20days%20immediately%20preceding%20GC%20contraction%20are%20characterized%20by%20a%20surge%20in%20the%20numbers%20of%20Foxp3-expressing%20T%20cells%2C%20arising%20at%20least%20in%20part%20from%20up-regulation%20of%20Foxp3%20by%20the%20TFH%20population.%20Gain-of-function%20experiments%20suggest%20that%20Foxp3%20expression%20by%20TFH%20cells%20may%20aid%20in%20the%20process%20of%20GC%20shutdown.%5CnGerminal%20centers%20%28GCs%29%20are%20the%20site%20of%20immunoglobulin%20somatic%20hypermutation%20and%20affinity%20maturation%2C%20processes%20essential%20to%20an%20effective%20antibody%20response.%20The%20formation%20of%20GCs%20has%20been%20studied%20in%20detail%2C%20but%20less%20is%20known%20about%20what%20leads%20to%20their%20regression%20and%20eventual%20termination%2C%20factors%20that%20ultimately%20limit%20the%20extent%20to%20which%20antibodies%20mature%20within%20a%20single%20reaction.%20We%20show%20that%20contraction%20of%20immunization-induced%20GCs%20is%20immediately%20preceded%20by%20an%20acute%20surge%20in%20GC-resident%20Foxp3%2B%20T%20cells%2C%20attributed%20at%20least%20partly%20to%20up-regulation%20of%20the%20transcription%20factor%20Foxp3%20by%20T%20follicular%20helper%20%28TFH%29%20cells.%20Ectopic%20expression%20of%20Foxp3%20in%20TFH%20cells%20is%20sufficient%20to%20decrease%20GC%20size%2C%20implicating%20the%20natural%20up-regulation%20of%20Foxp3%20by%20TFH%20cells%20as%20a%20potential%20regulator%20of%20GC%20lifetimes.%5CnImmunological%20T%20follicular%20helper%20cells%20that%20up-regulate%20the%20transcription%20factor%20Foxp3%20are%20required%20to%20turn%20off%20germinal%20center%20reactions.%5CnImmunological%20T%20follicular%20helper%20cells%20that%20up-regulate%20the%20transcription%20factor%20Foxp3%20are%20required%20to%20turn%20off%20germinal%20center%20reactions.%22%2C%22date%22%3A%222021%5C%2F07%5C%2F16%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1126%5C%2Fscience.abe5146%22%2C%22ISSN%22%3A%220036-8075%2C%201095-9203%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fscience.sciencemag.org%5C%2Fcontent%5C%2F373%5C%2F6552%5C%2Feabe5146%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-07-16T17%3A35%3A01Z%22%7D%7D%5D%7D
