to%20develop%20graft-versus-host%20disease%20after%20HSCT.%20Further%20work%20will%20need%20to%20determine%20whether%20Linc00402%20is%20a%20therapeutic%20candidate%20against%20allograft%20rejection%20or%20acute%20graft-versus-host%20disease.%5CnMechanisms%20governing%20allogeneic%20T%20cell%20responses%20after%20solid%20organ%20and%20allogeneic%20hematopoietic%20stem%20cell%20transplantation%20%28HSCT%29%20are%20incompletely%20understood.%20To%20identify%20lncRNAs%20that%20regulate%20human%20donor%20T%20cells%20after%20clinical%20HSCT%2C%20we%20performed%20RNA%20sequencing%20on%20T%20cells%20from%20healthy%20individuals%20and%20donor%20T%20cells%20from%20three%20different%20groups%20of%20HSCT%20recipients%20that%20differed%20in%20their%20degree%20of%20major%20histocompatibility%20complex%20%28MHC%29%20mismatch.%20We%20found%20that%20lncRNA%20differential%20expression%20was%20greatest%20in%20T%20cells%20after%20MHC-mismatched%20HSCT%20relative%20to%20T%20cells%20after%20either%20MHC-matched%20or%20autologous%20HSCT.%20Differential%20expression%20was%20validated%20in%20an%20independent%20patient%20cohort%20and%20in%20mixed%20lymphocyte%20reactions%20using%20ex%20vivo%20healthy%20human%20T%20cells.%20We%20identified%20Linc00402%2C%20an%20uncharacterized%20lncRNA%2C%20among%20the%20lncRNAs%20differentially%20expressed%20between%20the%20mismatched%20unrelated%20and%20matched%20unrelated%20donor%20T%20cells.%20We%20found%20that%20Linc00402%20was%20conserved%20and%20exhibited%20an%2088-fold%20increase%20in%20human%20T%20cells%20relative%20to%20all%20other%20samples%20in%20the%20FANTOM5%20database.%20Linc00402%20was%20also%20increased%20in%20donor%20T%20cells%20from%20patients%20who%20underwent%20allogeneic%20cardiac%20transplantation%20and%20in%20murine%20T%20cells.%20Linc00402%20was%20reduced%20in%20patients%20who%20subsequently%20developed%20acute%20graft-versus-host%20disease.%20Linc00402%20enhanced%20the%20activity%20of%20ERK1%20and%20ERK2%2C%20increased%20FOS%20nuclear%20accumulation%2C%20and%20augmented%20expression%20of%20interleukin-2%20and%20Egr-1%20after%20T%20cell%20receptor%20engagement.%20Functionally%2C%20Linc00402%20augmented%20the%20T%20cell%20proliferative%20response%20to%20an%20allogeneic%20stimulus%20but%20not%20to%20a%20nominal%20ovalbumin%20peptide%20antigen%20or%20polyclonal%20anti-CD3%5C%2FCD28%20stimulus.%20Thus%2C%20our%20studies%20identified%20Linc00402%20as%20a%20regulator%20of%20allogeneic%20T%20cell%20function.%5CnLncRNAs%20are%20differentially%20expressed%20by%20allogeneic%20antigen-stimulated%20T%20cells%2C%20and%20Linc00402%20regulates%20allogeneic%20T%20cells.%5CnLncRNAs%20are%20differentially%20expressed%20by%20allogeneic%20antigen-stimulated%20T%20cells%2C%20and%20Linc00402%20regulates%20allogeneic%20T%20cells.%22%2C%22date%22%3A%222021%5C%2F03%5C%2F17%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1126%5C%2Fscitranslmed.aaz0316%22%2C%22ISSN%22%3A%221946-6234%2C%201946-6242%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fstm.sciencemag.org%5C%2Fcontent%5C%2F13%5C%2F585%5C%2Feaaz0316%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-03-19T06%3A14%3A29Z%22%7D%7D%5D%7D
