3%29%20and%20granzyme%20B%20production%20by%20CD8%2B%20T%20cells.%5CnRATIONALEWe%20hypothesized%20that%20obtaining%20structural%20information%20for%20the%20complete%20IL-10%20receptor%20complex%20would%20enable%20the%20rational%20design%20of%20IL-10%20analogs%20with%20enhanced%20functional%20specificity%20and%20improved%20therapeutic%20utility.%20Mechanistically%2C%20IL-10%20functions%20as%20a%20secreted%20homodimer%20that%20engages%20two%20copies%20of%20a%20heterodimeric%20receptor%20complex%20comprising%20the%20private%20receptor%20subunit%2C%20IL-10R%5Cu03b1%2C%20and%20the%20shared%20subunit%2C%20IL-10R%5Cu03b2.%20The%20IL-10%5Cu2013dependent%20dimerization%20of%20IL-10R%5Cu03b1%20and%20IL-10R%5Cu03b2%20in%20turn%20initiates%20activation%20of%20the%20transcription%20factor%20STAT3%2C%20which%20mediates%20the%20diverse%20biological%20effects%20of%20IL-10.%20However%2C%20structural%20information%20for%20the%20complete%20IL-10%20receptor%20signaling%20complex%20is%20lacking%2C%20due%20in%20part%20to%20the%20extremely%20low%20affinity%20of%20IL-10%20for%20IL-10R%5Cu03b2.%5CnRESULTSTo%20overcome%20this%20limitation%2C%20we%20first%20used%20yeast%20display-based%20directed%20evolution%20to%20engineer%20a%20%5Cu201csuper-10%5Cu201d%20variant%20with%20greatly%20enhanced%20affinity%20for%20IL-10R%5Cu03b2%2C%20enabling%20assembly%20of%20the%20hexameric%20IL-10%5Cu2013IL-10R%5Cu03b1%5Cu2013IL-10R%5Cu03b2%20complex.%20Using%20this%20stabilized%20complex%2C%20we%20then%20determined%20the%20structure%20of%20the%20complete%20IL-10%20receptor%20complex%20at%203.5%20%5Cu00c5%20resolution%20by%20cryo%5Cu2013electron%20microscopy%20%28cryo-EM%29.%20The%20structure%20revealed%20how%20IL-10%20engages%20IL-10R%5Cu03b2%20to%20initiate%20signal%20transduction%2C%20and%20also%20uncovered%20the%20molecular%20basis%20for%20a%20mutation%20in%20IL-10R%5Cu03b2%20associated%20with%20early-onset%20IBD.%20In%20addition%2C%20the%20structure%20provided%20an%20engineering%20blueprint%20for%20the%20design%20of%20IL-10%20variants%20with%20which%20we%20could%20pharmacologically%20probe%20the%20nature%20of%20IL-10%5Cu2019s%20functional%20pleiotropy.Characterization%20of%20these%20IL-10%20variants%20revealed%20that%20the%20plasticity%20of%20IL-10%20signaling%20varies%20extensively%20across%20immune%20cell%20types%2C%20inversely%20correlating%20with%20the%20level%20of%20IL-10R%5Cu03b2%20expression.%20In%20particular%2C%20we%20found%20that%20myeloid%20cells%20exhibit%20robust%20STAT3%20activation%20in%20response%20to%20IL-10%20variants%20across%20a%20wide%20range%20of%20IL-10R%5Cu03b2%5Cu2013binding%20affinities%2C%20whereas%20IL-10%20signaling%20in%20lymphocytes%20was%20highly%20tunable.%20Several%20engineered%20IL-10%20variants%20exploited%20these%20differences%20to%20elicit%20myeloid-biased%20signaling%20in%20both%20cell%20lines%20and%20human%20peripheral%20blood%20mononuclear%20cells%20%28PBMCs%29.%20Functionally%2C%20these%20variants%20effectively%20inhibited%20inflammatory%20monocyte%20and%20macrophage%20activation%20in%20vivo%20and%20promoted%20survival%20in%20a%20mouse%20model%20of%20sepsis%2C%20thus%20retaining%20the%20major%20anti-inflammatory%20effects%20of%20IL-10.%20However%2C%20they%20showed%20significantly%20reduced%20capacity%20to%20stimulate%20proinflammatory%20gene%20expression%20in%20CD8%2B%20T%20cells%20and%20failed%20to%20potentiate%20IFN-%5Cu03b3%20or%20granzyme%20B%20production%2C%20thereby%20uncoupling%20the%20major%20opposing%20functions%20of%20IL-10.%5CnCONCLUSIONThe%20cryo-EM%20structure%20of%20the%20IL-10%20receptor%20complex%20yields%20key%20insights%20into%20the%20mechanisms%20of%20IL-10%20signaling%20and%20functional%20pleiotropy.%20Characterization%20of%20IL-10%5Cu2013derived%20partial%20agonists%20and%20super-agonists%20revealed%20that%20modulating%20receptor%20affinity%20can%20%5Cu201ctune%5Cu201d%20IL-10%20signaling%20in%20a%20cell%20type%5Cu2013selective%20manner%2C%20exploiting%20differences%20in%20IL-10%20response%20thresholds%20between%20immune%20cell%20populations.%20These%20results%20suggest%20a%20model%20in%20which%20the%20differential%20expression%20of%20the%20shared%20receptor%20subunit%20IL-10R%5Cu03b2%20results%20in%20distinct%20IL-10%20response%20thresholds%20across%20immune%20cell%20populations%2C%20enhancing%20the%20functional%20specificity%20of%20IL-10.%20By%20exploiting%20these%20natural%20features%20of%20IL-10%20signaling%2C%20we%20engineered%20IL-10%20variants%20exhibiting%20myeloid%20cell%20selectivity%2C%20which%20effectively%20uncoupled%20the%20pro-%20and%20anti-inflammatory%20functions%20of%20IL-10.%20These%20findings%20provide%20a%20conceptual%20framework%20for%20the%20development%20of%20improved%20cytokine-based%20therapeutics.%20%3Cimg%20class%3D%5C%22fragment-image%5C%22%20aria-describedby%3D%5C%22F1-caption%5C%22%20src%3D%5C%22https%3A%5C%2F%5C%2Fscience.sciencemag.org%5C%2Fcontent%5C%2Fsci%5C%2F371%5C%2F6535%5C%2Feabc8433%5C%2FF1.medium.gif%5C%22%5C%2F%3E%20Download%20high-res%20image
