20transcription%20factors%20present%20in%20nuclear%20lysate%20are%20bound%20to%20plasmid%20DNA%2C%20co-extracted%20and%20quantified%20using%20mass-spectrometry.%20The%20role%20of%20altered%20activity%20of%20transcription%20factor%20CtBP2%20was%20further%20examined%20by%20modification%20of%20its%20expression%20levels%20using%20CAG-rtTA3-CtBP2%20overexpression%20in%20small%20intestinal%20organoids%2C%20shCtBP2%20knockdown%20in%20LS174T%20cells%2C%20and%20familial%20adenomatous%20polyposis%20patient-derived%20organoids.%20CtBP2%20overexpression%20organoids%20were%20challenged%20by%20ER%20stress%20and%20ionizing%20irradiation.%20We%20identified%20a%20unique%20set%20of%20transcription%20factors%20with%20altered%20activation%20upon%20ER%20stress.%20Gene%20ontology%20analysis%20showed%20that%20transcription%20factors%20with%20diminished%20binding%20were%20involved%20in%20cellular%20differentiation%20processes.%20ER%20stress%20decreased%20CtBP2%20protein%20expression%20in%20mouse%20small%20intestine.%20ER%20stress%20induced%20loss%20of%20CtBP2%20expression%20which%20was%20rescued%20by%20inhibition%20of%20PERK%20signaling.%20CtBP2%20was%20overexpressed%20in%20mouse%20and%20human%20colorectal%20adenomas.%20Inducible%20CtBP2%20overexpression%20in%20organoids%20conferred%20higher%20clonogenic%20potential%2C%20resilience%20to%20irradiation-induced%20damage%20and%20a%20partial%20rescue%20of%20ER%20stress-induced%20loss%20of%20stemness.%20Using%20an%20unbiased%20proteomics%20approach%2C%20we%20identified%20a%20unique%20set%20of%20transcription%20factors%20for%20which%20DNA-binding%20activity%20is%20lost%20directly%20upon%20ER%20stress.%20We%20continued%20investigating%20the%20function%20of%20co-regulator%20CtBP2%2C%20and%20show%20that%20CtBP2%20mediates%20ER%20stress-induced%20loss%20of%20stemness%20which%20supports%20the%20intestinal%20stem%20cell%20state%20in%20homeostatic%20stem%20cells%20and%20colorectal%20cancer%20cells.%22%2C%22date%22%3A%222021-05-10%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41598-021-89326-w%22%2C%22ISSN%22%3A%222045-2322%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41598-021-89326-w%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-05-12T23%3A23%3A52Z%22%7D%7D%5D%7D
