s%20%28TCRs%29%20were%20identified%20by%20deep%20sequencing%20of%20gDNA%20isolated%20from%20T%20cells%2C%20and%20the%20TCR%20specificity%20was%20confirmed%20by%20transferring%20TCRs%20to%20the%20T%20cell%20line%20SKW-3%20or%20donor%20T%20cells.%5CnResults%20The%20co-culture%20was%20effective%20in%20the%20generation%20of%20CD8%20%2B%20or%20CD4%2BopT%20cells.%20The%20opT%20cells%20killed%20autologous%20tumors%20in%20a%20granzyme%20B%20or%20Fas-Fas%20ligand-dependent%20manner%20and%20expressed%20markers%20of%20tissue-resident%20memory%20phenotype.%20Each%20patient-derived%20opT%20cell%20culture%20displayed%20a%20unique%20complement%20of%20checkpoint%20proteins.%20Interestingly%2C%20only%20NKG2A%20blockade%20showed%20a%20potent%20increase%20in%20the%20interferon-%5Cu03b3%20production%20compared%20with%20blocking%20programmed%20cell%20death%20protein%201%20%28PD-1%29%20or%20programmed%20cell%20death%20ligand%201%20%28PD-L1%29%20or%20TIM3%20or%20TIGIT%20or%20LAG3.%20Importantly%2C%20TCR%20sequencing%20demonstrated%20a%20dramatic%20clonal%20expansion%20of%20T%20cells%20with%20a%20restricted%20subset%20of%20TCRs.%20Cloning%20and%20transferring%20the%20TCRs%20to%20heterologous%20T%20cells%20was%20sufficient%20to%20confer%20tumor%20cell%20recognition%20and%20cytotoxic%20properties%20in%20a%20patient-specific%20manner.%5CnConclusion%20We%20report%20a%20platform%20for%20expanding%20tumor-targeting%20T%20cells%20from%20the%20peripheral%20blood%20of%20patients%20with%20pancreatic%20cancer.%20We%20identify%20the%20NKG2A-HLA-E%20axis%20as%20a%20potentially%20important%20checkpoint%20for%20CD8%20%2BT%20cells%20for%20pancreatic%20cancer.%20Lastly%2C%20we%20demonstrate%20empirical%20identification%20of%20tumor-targeting%20TCRs%20that%20can%20be%20used%20for%20TCR-therapeutics.%22%2C%22date%22%3A%222021%5C%2F11%5C%2F01%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1136%5C%2Fjitc-2021-003213%22%2C%22ISSN%22%3A%222051-1426%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fjitc.bmj.com%5C%2Fcontent%5C%2F9%5C%2F11%5C%2Fe003213%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-11-22T18%3A41%3A55Z%22%7D%7D%5D%7D
