%22Macrophages%20have%20a%20key%20role%20in%20shaping%20the%20tumour%20microenvironment%20%28TME%29%2C%20tumour%20immunity%20and%20response%20to%20immunotherapy%2C%20which%20makes%20them%20an%20important%20target%20for%20cancer%20treatment1%2C2.%20However%2C%20modulating%20macrophages%20has%20proved%20extremely%20difficult%2C%20as%20we%20still%20lack%20a%20complete%20understanding%20of%20the%20molecular%20and%20functional%20diversity%20of%20the%20tumour%20macrophage%20compartment.%20Macrophages%20arise%20from%20two%20distinct%20lineages.%20Tissue-resident%20macrophages%20self-renew%20locally%2C%20independent%20of%20adult%20haematopoiesis3%5Cu20135%2C%20whereas%20short-lived%20monocyte-derived%20macrophages%20arise%20from%20adult%20haematopoietic%20stem%20cells%2C%20and%20accumulate%20mostly%20in%20inflamed%20lesions1.%20How%20these%20macrophage%20lineages%20contribute%20to%20the%20TME%20and%20cancer%20progression%20remains%20unclear.%20To%20explore%20the%20diversity%20of%20the%20macrophage%20compartment%20in%20human%20non-small%20cell%20lung%20carcinoma%20%28NSCLC%29%20lesions%2C%20here%20we%20performed%20single-cell%20RNA%20sequencing%20of%5Cu00a0tumour-associated%20leukocytes.%20We%20identified%20distinct%20populations%20of%20macrophages%20that%20were%20enriched%20in%20human%20and%20mouse%20lung%20tumours.%20Using%20lineage%20tracing%2C%20we%20discovered%20that%20these%20macrophage%20populations%20differ%20in%20origin%20and%20have%20a%20distinct%20temporal%20and%20spatial%20distribution%20in%20the%20TME.%20Tissue-resident%20macrophages%20accumulate%20close%20to%20tumour%20cells%20early%20during%20tumour%20formation%20to%20promote%20epithelial%5Cu2013mesenchymal%20transition%20and%20invasiveness%20in%20tumour%20cells%2C%20and%20they%20also%20induce%20a%20potent%20regulatory%20T%20cell%20response%20that%20protects%20tumour%20cells%20from%20adaptive%20immunity.%20Depletion%20of%20tissue-resident%20macrophages%20reduced%20the%20numbers%20and%20altered%20the%20phenotype%20of%20regulatory%20T%20cells%2C%20promoted%20the%20accumulation%20of%20CD8%2B%20T%20cells%20and%20reduced%20tumour%20invasiveness%20and%20growth.%20During%20tumour%20growth%2C%20tissue-resident%20macrophages%20became%20redistributed%20at%20the%20periphery%20of%20the%20TME%2C%20which%20becomes%20dominated%20by%20monocyte-derived%20macrophages%20in%20both%20mouse%20and%20human%20NSCLC.%20This%20study%20identifies%20the%20contribution%20of%20tissue-resident%20macrophages%20to%20early%20lung%20cancer%20and%20establishes%20them%20as%20a%20target%20for%20the%20prevention%20and%20treatment%20of%20early%20lung%20cancer%20lesions.%22%2C%22date%22%3A%222021-06-16%22%2C%22language%22%3A%22en%22%2C%22DOI%22%3A%2210.1038%5C%2Fs41586-021-03651-8%22%2C%22ISSN%22%3A%221476-4687%22%2C%22url%22%3A%22https%3A%5C%2F%5C%2Fwww.nature.com%5C%2Farticles%5C%2Fs41586-021-03651-8%22%2C%22collections%22%3A%5B%5D%2C%22dateModified%22%3A%222021-06-16T20%3A04%3A39Z%22%7D%7D%5D%7D
